How much equity to ask for? I’m new… Them ~2yrs

I’ve caught the interest of an early stage, pre-funded startup, small team testing an MVP. They’ve been at it for almost two years, almost one year full time. I bring a lot of value, domain expertise, startup experience and the founders are interested in me. I was introduced by a trusted source  – so the trust element is there too. 

I can only work part time for now while I am currently employed full time. How much equity should I ask for / expect? I am expecting they bring me on as a first employee potentially in a senior leadership role. Best part is, I work 2 blocks from them in DTLA

It depends entirely on how much value you add and how likely the company is to succeed without you. For a pre-revenue, pre-funding sweat equity position your equity stake should be much higher than an employee #1 would normally get. You should probably accept any reasonable vesting schedule, to compensate for the fact that they've been working on the startup for a long time already.
